چکیده

Eremothecium ashbyii is one of a number of organisms which produce considerable amounts of riboflavin, most of which is secreted by the cells into the medium (Guilliermond, 1935). The investigations of Schopfer (1944) and Schopfer and Guilloud (1945) resulted in the development of a synthetic medium for this organism which permitted growth and riboflavin production to occur under chemically defined conditions. Subsequent studies by Yaw (1948) have led to a modification of the original medium to iiZclude the amino acids, methionine and histidine. With the determination of the nutritional requirements of E. ashbyii, it became possible to undertake studies concerning the mechanism of riboflavin biosynthesis by means of precursors which modify riboflavin yield. Such an approach was utilized by Yaw (1948), who tested certain synthetic compounds for precursor activity. However, none of the compounds tested was found to be active in the biosynthesis of riboflavin. The present study is concerned with the purines and pyrimidines in the biosynthesis of riboflavin by E. ashbyii. These compounds were selected for the following reasons: (1) There are marked similarities in chemical structure among the purines, pyrimidines, and riboflavin, all of which contain a diazine ring, and (2) the purines and pyrimidines are naturally occurring compoun$ds. It was therefore hoped that the information obtained would provide a better understanding of riboflavin biosynthesis, about which very little is known at present (Williams et al., 1950).

Studies on the synthesis of riboflavin by a mutant yeast, Saccharomyces cerevisiae.

Though the synthesis of riboflavin in large amounts by microorganisms has been encountered, very little is known about the mechani involved in the biosynthesis of this vitamin. A critical examination of the various environmental conditions for different organisms producing this vitamin showed that the optimum conditions for the production of riboflavin varied widely from organism to organism (S...

Development of specific experimental systems for flavinogenesis using non-growing cell of Eremothecium ashbyii.

Experimental systems using non-growing cell of Eremothecium ashbyii to elucidate the mechanism of flavinogenesis were established in this paper. 1. It was found that vacuum infiltration of purines for 2, 3 and 5min and moderate shaking of non-growing cell brought about a good reproducibility of the stimulatory effects on flavinogenesis by purines and furthermore, a higher flavinogenesis. 2. The...

The utilization of the ureide carbon of adenine in the biosynthesis of riboflavin in Eremothecium ashbyii.

There is considerable evidence that a critical reaction in purine synthesis is the closure of the purine ring by the addition of a “formate” group to 4-amino-5-imidazolecarboxamide ribonucleotide (1). Apparently the “formate” is added as an active intermediate, since formate as such has little effect in stimulating purine synthesis. Indeed, there is a demonstrable difference in the utilization ...

Some nutritional requirements for biosynthesis of riboflavin by Eremothecium ashbyii.

Eremothecium ashbyii Guilliermond is a remarkable organism because of its capability of synthesizing relatively large amounts of riboflavin under appropriate conditions. This vitamin is synthesized to a limited extent by a rather wide variety of microorganisms (Pridham, 1952), but those known particularly for their riboflavin producing abilities along with E. a&hbyii are Ashbya gosypi'i (Wicker...

The incorporation of the pyrimidine ring of adenine into the isoalloxazine ring of riboflavin.

In the biogenesis of riboflavin by the yeast, Eremothecium ashbyii, carbon atoms constituting the pyrimidine ring of adenine were incorporated into the 6,7-dimethylisoalloxazine ring of riboflavin with much greater efficiency than was the 8 position of the purine ring (1). It remained to determine whether the carbon atoms which constitute the pyrimidine ring of the purine are incorporated direc...
